چکیده

A sulphamidase and a sulphoesterase were isolated from adapted cells of Flavobacterium heparinum. These enzymes were partially purified from the 'heparinases' present in the bacterial extracts and characterized. The sulphamidase has a high specificity for glucosamine N-sulphate and glucosamine 2,6-disulphate. The activity decreases sharply with increasing molecular weight ofthe substrates tested. The sulphamidase and the sulphoesterase activities were distinguished from each other by their different sensitivities to concentration of phosphate ion and to temperature. The importance of these enzymes in the study of the structure of heparin is discussed.

High-density-lipoprotein cholesterol in heparin-MnCl2 supernates determined with the Dow enzymic method after precipitation of Mn2+ with HCO3-.

Manganese interferes with enzymic cholesterol methods. In this study, we enzymically measured high-density-lipoprotein (HDL) cholesterol in heparin-Mn2+ supernates that had been treated with NaHCO3 (91 mmol/L) to precipitate Mn2+, and compared results with those by an automated Liebermann- Burchard method. For untreated supernates of 96 fresh plasma samples, the enzymic values were 10.4% higher...

Expression in Escherichia coli, purification and characterization of heparinase I from Flavobacterium heparinum.

The use of heparin for extracorporeal therapies has been problematical due to haemorrhagic complications; as a consequence, heparinase I from Flavobacterium heparinum is used for the determination of plasma heparin and for elimination of heparin from circulation. Here we report the expression of recombinant heparinase I in Escherichia coli, purification to homogeneity and characterization of th...

Inactivation of lipoprotein lipase by heparinase.

The ability of administered heparin to “clear” lipemic plasma has been adequately explained (2, 3) in that it induces the appearance in the plasma of lipoprotein lipase, an enzyme which catalyzes the hydrolysis of the triglyceride moiety of chylomicrons and &lipoproteins. This has caused a shift in emphasis from heparin to the lipase which, normally present in several tissues (4), appears to pl...

Interaction of size-fractionated heparins with lipoprotein lipase and hepatic lipase in the rat.

Heparin and heparin partially depolymerized by enzymic digestion were separated into six size fractions. Hep 1 (tetrasaccharides), with a mean M(r) of 1200, did not release significant amounts of either lipoprotein lipase (LPL) or hepatic lipase (HL) on intravenous injection into rats. Hep 2 (mainly octa- and deca-saccharides), with a mean M(r) of 2400-3000, released both lipases. To evoke the ...

Heparin--magnesium chloride--albumin method for enzymic measurement of cholesterol in high-density lipoprotein.

We describe a new method for measuring high-density-lipoprotein cholesterol in sera, with use of heparin, MgCl2, and albumin to precipitate very-low- and low-density lipoproteins, Lipoprotein electrophoresis and immunochemical methods show that high-density lipoprotein is completely and specifically isolated in the supernate, the cholesterol concentration of which is estimated by the enzymic me...
